چکیده

Evidence-based medicine encourages the use of quantitative diagnostic test results to estimate the probability of a particular diagnosis. Likelihood ratios (LRs) are among the best tools for maximizing the diagnostic information gained from diagnostic assays that provide results on a continuous scale. They provide the odds that an animal with a particular test result actually has the disease in question based on the magnitude of the test result. A commercial enzyme-linked immunosorbent assay (ELISA) was used to test sera from 143 dairy cattle infected with Mycobacterium paratuberculosis and 2974 cattle free of this infection. This assay transforms ELISA reader optical density values into sample-to-positive (S/P) ratios. The LR was calculated for S/P results from 0.00 to 1.00 at 0.05-S/P unit intervals. LRs were directly but not linearly correlated with ELISA S/P ratios (r(2) = 0.94). The mathematical function describing the relationship between the ELISA S/P ratio and the LR was LR = 265 x (S/P value)(2.03). LRs were also directly related to the frequency of animals testing positive for paratuberculosis by fecal culture and other serologic tests. Based on these LRs, guidelines for interpretation and application of this ELISA for the diagnosis and control of paratuberculosis in dairy cattle herds are recommended.

Evaluation of four commercial enzyme-linked immunosorbent assays for the diagnosis of bovine paratuberculosis in Chilean dairy herds.

The accuracy of 4 commercial enzyme-linked immunosorbent assays (ELISAs) for diagnosis of bovine paratuberculosis was compared using sera from 53 Mycobacterium avium subsp. paratuberculosis (MAP) fecal culture-positive dairy cows (cases) and sera from 345 dairy cattle resident in 11 fecal culture-negative herds on 2 consecutive occasions 1 year apart (controls). The specificity of all 4 ELISA k...

Diagnosis of bovine paratuberculosis by a novel enzyme-linked immunosorbent assay based on early secreted antigens of Mycobacterium avium subsp. paratuberculosis.

We previously reported that protein antigens of serodiagnostic potential were more abundant in culture filtrates than cellular extracts from liquid cultures of Mycobacterium avium subsp. paratuberculosis (D. Cho and M. T. Collins, Clin. Vaccine Immunol. 13:1155-1161, 2006). Based on this observation, a novel enzyme-linked immunosorbent assay (ELISA) using antigens secreted by young (early- to m...

AhpC, AhpD, and a secreted 14-kilodalton antigen from Mycobacterium avium subsp. paratuberculosis distinguish between paratuberculosis and bovine tuberculosis in an enzyme-linked immunosorbent assay.

Sera from cattle naturally infected with Mycobacterium avium subsp. paratuberculosis (n = 56) and naturally (n = 4) and experimentally (n = 8) infected with Mycobacterium bovis were tested for the presence of antibodies against paratuberculosis antigens. An enzyme-linked immunosorbent assay (ELISA) was established based on absorption of M. avium subsp. paratuberculosis antigens on a hyperimmune...

Adaptation of an indirect enzyme-linked immunosorbent assay by purified gp51SU for detection of antibodies to bovine leukemia virus

The objective of this study was to compare an indirect ELISA, based on a purified 60 kDa envelope glycoprotein (gp51SU), with a Pourquire indirect ELISA for the detection of antibodies to the bovine leukemia virus. For conducting this research, 340 serum samples were collected from two different breeds of cows (Sarabi and Holestin) in different herds. Commercial ELISA revealed positive results ...

DEVELOPMENT OF A SIMPLE AND SENSITIVE ENZYME- LINKED IMMUNOSORBENT ASSAY (ELISA) FOR CLINICAL MEASUREMENT OF TESTOSTERONE USING PENICILLINASE AS LABEL

An enzyme-linked immunosorbent assay using a homologous combination of antiserum raised against testosterone-3-0-carboxymethyloxime-bovine serum albumin (T-3-0-CMO-BSA ) and penicillinase-labelled T-3-0-CMO was developed. This assay was utilized to measure testosterone in serum samples of male and female subjects. The sensitivity of the assay is 50pg/well and the antibody developed crossrea...
